How to set it in a single-player game?

Plugin for Spigot/Paper servers
The previous method is only applicable for Vanilla Minecraft. If you’re using a modified server on Spigot/Paper, you’ll need a plugin, such as EssentialsX. Install this and restart the game world.
Then, users only need to write the command in chat: /set spawn. Done! The respawn point is made. Players can teleport to the desired location using the /spawn command. This is not the only plugin available. Each user will be able to find a more suitable option.
Mods for Forge/Fabric
If a server does not support certain plugins, modifications are always available to help.
